Orf virus (ORFV), a member of parapoxvirus, is an enveloped virus with genome
of double-stranded DNA. ORFV causes contagious pustular dermatitis or contagious ecthyma
in sheep and goats worldwide. In general, detection of viral DNA and observing ORFV virion
in tissues of afflicted animals are two methods commonly used for diagnosis of orf
infection; however, isolation of the ORFV in cell culture using virus-containing tissue as
inoculum is known to be difficult. In this work, the ORFV (Hoping strain) isolated in
central Taiwan was successfully grown in cell culture. We further examined the biochemical
characteristic of our isolate, including viral genotyping, viral mRNA and protein
expression. By electron microscopy, one unique form of viral particle from ORFV infected
cellular lysate was demonstrated in the negative-stained field. Moreover, immunomodulating
and anti-influenza virus properties of this ORFV were investigated. ORFV stimulated human
monocytes (THP-1) secreting proinflammatory cytokines IL-8 and TNF-α. And, pre-treatment
of ORFV-infected cell medium prevents A549 cells from subsequent type A influenza virus
(IAV) infection. Similarly, mice infected with ORFV via both intramuscular and
subcutaneous routes at two days prior to IAV infection significantly decreased the
replication of IAV. In summary, the results of a current study indicated our Hoping strain
harbors the immune modulator property; with such a bio-adjuvanticity, we further proved
that pre-exposure of ORFV protects animals from subsequent IAV infection. 相似文献

Newcastle disease virus (NDV) causes a highly contagious viral disease in poultry and
wild birds, and it can cause significant economic loss worldwide. Eight viral strains were
isolated by inoculating embryonated chicken eggs from the Poyang Lake region of China with
swab samples. All eight of the NDV isolates were identified as class I genotype 3 strains,
but they diverged notablely from class II viruses. Further analysis revealed that all
eight NDV isolates were lentogenic strains containing the
112ERQER↓L117 motif at the F protein cleavage site. The strains
were highly identical and were more species specific (chicken and waterfowl) than site
specific (Nanchang and Duchang regions). The close phylogenetic proximity of these
isolates indicates that viral transmission may happen between poultry and wild birds. Our
study demonstrates that lentogenic class I NDVs exist in clinically healthy wild waterfowl
and poultry within the Poyang Lake region. Active surveillance of these viruses to
determine their evolution and origin is one of the most realistic strategies for

To clarify the pathogenicity of Japanese type 1 porcine reproductive and respiratory
syndrome virus (PRRSV) isolate in experimentally infected pigs, we evaluated clinical
signs and monitored viremia for 21 days post-inoculation (dpi). Lungs were mottled, tanned
and reddish in appearance; had lesions predominantly in the cranial, middle and accessory
lobes; and failed to collapse at 10 dpi. Although microscopic lesions of lungs were
reproduced using the Japanese emerging type 1 PRRSV isolate under experimental conditions,
no significant differences were noted between the challenge and control groups regarding
mean rectal temperature and daily weight gain. These results provide useful insights into
the limited pathogenicity of single infection with the Japanese type 1 PRRSV isolate in

The authors analysed epidemiological data of the Hungarian tick-borne encephalitis epidemic from the past seven decades. A total of 911 meningitis serosa cases were described from 1930-1950 s by local hospital physicians, indicating that the virus had been present in the country decades before its official identification in 1952. The virus spread freely in the 1950s–1960s, occupying almost all habitats where ticks occurred in large numbers. The increasing number of cases drove authorities to classify this illness as a notifiable disease in 1977 and to organize the first measures to stop the epidemic. Statistical analysis revealed that the large-scale vaccination launched from the 1990s was responsible for the sharp decrease in the number of human cases from 1997. A significant negative correlation was found between the number of vaccine doses sold and human cases 6 years later. The TBEV endemic area covers 16.57% of the territory and 16.65% of the population of the country. In the last 10 years, 186,000 vaccine doses/year in average were enough to keep the incidence of human TBEV infections between 0.45 and 0.06/100,000 persons. A 20-year-long study found evidence for easing clinical signs in TBEV-infected hospitalized patients. Statistics found a sharp decrease in the number of samples sent for TBEV diagnosis after 1989. Male dominance of patients was characteristic of the epidemics since the 1940s, but now analysis of detailed data from the 1981–2021 period (60.5%–87.5%) proved the statistical significance of this dominance. Obviously, the voluntary vaccination programme was the tool which broke the spread of the epidemic. Widespread public awareness of the disease and the tick vector, probable evolutionary spread of less pathogenic virus strains supplemented with the vaccination campaign led to a negligible level of human TBE cases in Hungary in the last years. 相似文献

Surveillance of hepatitis E virus (HEV) in risk groups is an important strategy to monitor its circulation pattern and to timely detect changes thereof. The aims of this cross-sectional study were to estimate the prevalence of HEV infections in pigs and humans from different regions of the country, to identify risk factors for increasing anti-HEV IgG prevalence and to characterize HEV strains. The presence of anti-HEV antibodies was assessed by commercial ELISA in serum samples from the general population, farm and slaughterhouse employees, as well as pigs sampled in the three regions of Cuba from February to September 2016. Overall, individuals with occupational exposure to swine or swine products (70/248, 28.2%) were 4 times more likely to be seropositive compared to the general population (25/285, 8.7%; OR: 4.18; p < .001). Within the risk group, risk factors included age, number of years working in a professional activity with direct exposure to swine, geographic region and distance between residence and closest professional swine setting, while wearing gloves had a protective effect. Prevalence of total anti-HEV antibodies in swine was 88.2% (165/187) and HEV RNA was detected by real-time RT-PCR in 9.2% (16/173) swine stools. All HEV strains sequenced clustered within genotype 3. Some strains clearly belonged to subtype 3a, while another group of strains was related with subtypes 3b and 3 k but partial HEV sequences did not allow unequivocal subtype assignment. These findings suggest that the high HEV exposure in Cuban individuals with swine-related occupations could be due to enzootic HEV in certain regions, direct contact with infectious animals or their products as well as environmental contamination. 相似文献

为建立检测甜瓜黄斑病毒(melon yellow spot virus, MYSV)的SYBR Green Ⅰ实时荧光定量PCR(qPCR)方法。基于MYSV核衣壳蛋白基因保守序列设计qPCR特异性引物对,针对引物退火温度、引物浓度、特异性和敏感性进行系列优化。结果显示,优化后的qPCR方法最适退火温度为61.3℃,最适引物浓度为0.65μmol·L-1,特异性强,灵敏度高,比PCR高100倍。以携带目的基因片段的重组质粒为标准品,构建的qPCR标准曲线循环阈值与模板浓度呈良好的线性关系,相关系数为0.999 7。实验样品验证表明建立的qPCR方法可用于MYSV的定量检测。 相似文献
